Effect of Genetic Modification of Catalase Activity on the Dormancy and the Sprouting of Potato Mini Tubers (Solanum Tuberosum L.)
نویسنده
چکیده
In several vegetable species a relationship between the metabolism of active oxygen species and breakage of dormancy has been established. However, scarce information is available on the role of oxidative stress in potato tubers. The purpose of this investigation is to study the effect of CAT genetic modification on the dormancy break of potato tubers. A transgenic approach with partial repression of CAT activity in cv. Desirée was applied. The genes coding for the CAT 2 of Nicotiana plumbaginifolia (Cat 2AS) and of Gossypium hirsutum (GH) were used according to "antisense" and "co-suppression" strategies. The repression of CAT activity was associated with Ascorbate peroxidase (APX) accumulation and a decrease in the Hydrogen peroxide (H2O2) in potato tubers. The sprouting of transgenic tubers was accelerated as compared to the non-transformed control and was associated with a decrease of CAT activity and H2O2 contents. Keywords— Catalase, Dormancy, Hydrogen peroxide, Solanum tuberosum L. Abbreviations: EDTA: (Ethylene-diamine-tetra-acetic); H2O2: Hydrogen peroxide; NBT: (Nitroblue tetrazolium); SDS: (Sodium dodecyl sulphate); PAGE: (Polyacrylamide gel); PVPP: (Polyvinylpolypyrrolidone); TEMED: (Tetramethyl–ethylen-diamine).

Physiological and Biochemical Regulation of Dormancy and Sprouting in Potato Tubers (Solanum tuberosum L.)
At harvest, potato (Solanum tuberosum L.) tubers are in a dormant state and do not have the ability to sprout. After harvesting, tuber dormancy is released progressively. This release is accompanied by numerous physiological and biochemical changes, such as carbohydrate hydrolysis and the accumulation of saccharides, which are detrimental to the nutritional and processing qualities of potatoes....

To evaluate the effects of heat treatments on sprouting weight, loss and other charactristics of two potato cultivars (Agria and Diamont) in cold (9±1) and ambient (18±1) storage, a series of experiments were conducted in 2004. In this study, after curing, the tubers of both cultivars were treated with warm air, at 25, 35, 45 and 57oC for 4, 8 and 12 h and with warm water at 22, 54 and 57 for 1...

Optimization of mineral nutrition in aeroponic systems is one of the most significant factors in producing high-quality and high yields of healthy seed tubers in potato (Solanum tuberosum L.). The present study was conducted at the Research Farm and Greenhouse of Agricultural and Natural Resources Center of Ardabil during two successive years (2018-2019). For the aeroponic system an experiment ...
متن کاملDual effects of ethylene on potato dormancy and sprout growth.
Dormant potato tubers (Solanum tuberosum L.) of two cultivars were treated with various concentrations of ethylene gas for various exposure periods. As has been shown by others, ethylene caused a rapid but transient increase in respiration rate, which appeared to be independent of any effects on dormancy. All concentrations tested caused accelerated sprouting, 2 microliters per liter being the ...

In addition to common vegetative multiplication, potato crop can be reproduced by true potato seed (TPS). Only the limited number of pests and diseases, which are the most important causes of a decrease in potato production, can be transmitted by true potato seed. Hence, to evaluate the potential of true potato seed in comparison to potato seed tuber and mini tuber, an experiment was conducted ...
